DESCRIPTION NS 11394 is a drug with selectivity for the α3 and α5 subtypes. It acts as a subtype-selective positive allosteric modulator at GABAA receptors. It is used as an analgesic for use in chronic or neuropathic pain. It is unique in having superior efficacy at alpha(3) receptors and low efficacy at alpha(1) receptors in vitro. It has an excellent pharmacokinetic profile, which correlates with pharmacodynamic endpoints, yielding a high level of confidence in deriving in vivo conclusions anchored to an in vitro selectivity profile and allowing for translation to higher species. It impairs memory in both rats and mice because of its efficacy at GABA(A)-alpha(5) receptors. It is potent and highly effective in rodent anxiety models.
